Dual signal amplification coupling with DNA-templated silver nanoclusters for sensitive and label-free detection of thrombin

Abstract Sensitive and reliable determination of thrombin is relevant in the realms medical biological research as it serves an essential biomarker a number blood-related illnesses. Herein, we integrate allosteric probe-based specific identification dual signal amplification to present unique fluorescent technique for label-free sensitive detection. Based on DNA polymerase endonuclease-assisted amplification, method exhibits high sensitivity with low limit detection 2.3 pM, while maintaining excellent selectivity stability. More importantly, approach successfully applied analyzing effect nalbuphine coagulation function mice. Overall, this possesses advantages specificity thrombin, which promising diagnosis diseases.

Novel luminescent silver nanoclusters (AgNCs) were synthesized utilizing DNA as templates by a simple, rapid and one-pot procedure. Luminescence studies indicated that these DNA-AgNCs exhibited strong emission with peak maximum at 624 nm. The fluorescence of the DNA-AgNCs was found to be quenched by Cu(2+) enabling its detection with high sensitivity and selectivity.

A label-free double amplification system has been developed by using a ternary DNA probe containing the poly(adenine-thymine) sequence assisted by exonuclease III degradation. The method achieved more than 600-fold signal amplification and allowed sensitive detection of single-stranded DNA and thrombin at the pM level by using liquid chromatography/mass spectrometry.
